Despite the success of the clinical use of antibodies against EGFR in patients with colorectal cancer and squamous cell carcinoma of the head and neck, their low effectiveness in glioblastoma has been shown. Therefore, for the treatment of gliomas, a specific EGFR drug is needed, capable of penetrating into the tumor focus in the brain, and having low immunogenicity.</p> <p>In this work, aptamers – single-stranded DNA oligonucleotides specific to EGFR, U2 and Gol1 are presented as such a preparation. In this study, we obtained a cellular model of human glioma with EGFR and EGFRvIII overexpression, which showed the specificity of U2 and Gol1 aptamers to these receptors using classical methods, as well as the method of aptaimmunocytochemistry. A study of the effect of binding of the Gol1 aptamer to the EGFRvIII receptor on the next steps of the signaling pathway showed a change in the expression levels of genes associated with cell proliferation and survival (<italic>JUN</italic>, <italic>FOS</italic>, <italic>CCND1</italic>, <italic>PI3K</italic> and <italic>AKT3</italic>), while the U2 aptamer did not demonstrate a significant effect on cells <italic>in vitro</italic>.</p> <p>These results showed that the Gol1 aptamer has therapeutic potential against human glioblastoma tumor cells overexpressing the EGFRvIII mutant type receptor.</p></abstract><trans-abstract xml:lang="ru"><p>Гиперэкспрессия рецептора эпидермального фактора роста (EGFR) или его мутации опосредуют сигнальные пути, приводящие к пролиферации, инвазии опухолевых клеток, а также к повышению их выживаемости. Несмотря на успешность клинического применения антител против EGFR у пациентов с колоректальным раком и плоскоклеточным раком головы и шеи, показана их низкая эффективность при глиобластоме. Поэтому для терапии глиом необходим специфичный к EGFR препарат, способный проникнуть в опухолевый очаг в головном мозге и обладающий низкой иммуногенностью. В данной работе в качестве такого препарата представлены аптамеры – одноцепочечные ДНК-олигонуклеотиды, специфичные к EGFR, U2 и Gol1. В этом исследовании мы получили клеточную модель глиомы человека с гиперэкспрессией <italic>EGFR</italic> и <italic>EGFRvIII</italic>, на которой показали специфичность аптамеров U2 и Gol1 к данным рецепторам, используя классические методы, а также метод апта-иммуноцитохимии. Исследование влияния связывания аптамера Gol1 с рецептором <italic>EGFRvIII</italic> на следующие ступени сигнального пути показало изменение уровней экспрессии генов, связанных с пролиферацией и выживаемостью клеток (<italic>JUN, FOS, CCND1, PI3K и AKT3</italic>), в то время как аптамер U2 не продемонстрировал значимый эффект на клетках <italic>in vitro</italic>.
