Skin fibroblasts (n=5) were used for reference. The primary cultures were obtained by the method of explants. The experimental biofilm matrix included <italic>S. aureus, E. faecalis, A. baumannii, P. aeruginosa, K. pneumoniae, P. mirabilis</italic> (5 of each species).</p> <p><bold>Results. </bold>The elevated levels of TGF-1β and VEGF, secreted by the primary cultures of fibroblasts, against of the absence of changes in the values of FGF and GM-CSF, are the criteria for the imbalance of growth factors characteristic of CW. The impact of the biofilm matrix <italic>P. aeruginosa, K. pneumoniae, A. baumannii, P. mirabilis</italic> on skin fibroblasts was accompanied by an increase in the production of VEGF and GM-CSF and a decrease in the synthesis of TGF-1β and FGF. In the experiments with <italic>S. aureus</italic> and <italic>E. faecalis</italic>, a similar direction of changes was recorded, but the degree of its severity was lower than for experiments with the gram-negative bacteria.</p> <p><bold>Conclusion.</bold> The obtained results have demonstrated various effects of the biofilm matrix on the production of growth factors by fibroblasts and updated the information on the pathogenesis of CW formation. The revealed imbalance in the synthesis of growth factors by fibroblasts in the granulation tissue of wounds with “borderline” lifetimes (22–28 days) determines the need for tactical approaches in the treatment of CW.</p></abstract><trans-abstract xml:lang="ru"><p><bold>Цель</bold> – проанализировать продукцию фибробластами факторов роста в условиях раневого процесса и при воздействии матрикса бактериальной биопленки.</p> <p><bold>Материал и методы</bold>. Оценивали уровни факторов роста: VEGF, TGF-1β, GM-CSF, FGF, продуцируемых фибробластами грануляционной ткани ОР (группа 1, n=9) и ХР (группа 2, n=17). Для сопоставления данных использовались фибробласты кожи (n=5). Первичные культуры получали методом эксплантатов. В эксперименте применяли матрикс биопленки <italic>S. aureus, E. faecalis, A. baumannii, P. aeruginosa, K. pneumoniae, P. mirabilis</italic> (по 5 каждого вида).</p> <p><bold>Результаты.</bold> Повышенные уровни TGF-1β и VEGF, секретируемые первичными культурами фибробластов, на фоне отсутствия изменений со стороны значений FGF и GM-CSF, являются критериями дисбаланса факторов роста, характерного для ХР. Воздействие матрикса биопленки <italic>P. aeruginosa, K. pneumoniae, A. baumannii, P. mirabilis</italic> на фибробласты кожи сопровождалось увеличением продукции VEGF и GM-CSF, снижением синтеза TGF-1β и FGF. В опытах с<italic> S. aureus</italic> и <italic>E. faecalis</italic> регистрировалась аналогичная направленность изменений, однако степень ее выраженности была ниже, чем для опытов с грамотрицательными бактериями.</p> <p><bold>Заключение.</bold> Полученные результаты демонстрируют различные эффекты матрикса биопленки на выработку фибробластами факторов роста и дополняют информацию о патогенезе формирования ХР.
